So i am looking into buying a western digital 1Tb portable hard drive but just wondering if i can use it as my main hard drive.
I have a laptop with like 10GB out of 218 left.

yes you can you just need to format it to the right file type fat32 or ntfs more likely the later
put it in to the laptop and use your os disk to start instaling then use the hardware from the portable case to copy your data from your old hard drive

I have one 1TB USB 3.0 portable hard drive. I have 3 laptops on 2 pcs, can I use one hard drive to back up all 5 computers? Or do I need 5 separate hard drives? Thank you for your attention.

Providing you have sufficient space then no problem. You can do it without partitioning the drive by making five folders. One for each computers backup.
